Recap securely and allow 5 minutes at room temperature for rehydration. Aseptically transfer 25g of PT item matrix from the PT item container 2026MM01R1-1 to the enrichment broth for Salmonella. Transfer entire of the rehydrated content from vial 2026MM01R1-V1 to the enrichment broth for Salmonella ensuring thorough homogenization. Proceed with the routine analysis. Vial 2026MM01R1-V2 Uncap the PT item vial 2026MM01R1-V2 aseptically and rehydrate using 4ml sterile saline peptone water (0.1% Peptone with 0.85%NaCl). Recap securely and allow 5 minutes at room temperature for rehydration. Aseptically transfer 25g of PT item matrix from the PT item container 2026MM01R1-2 to the enrichment broth for Salmonella. Vial 2026MM01R1-V3 Uncap the PT item vial 2026MM01R1-V3 aseptically and rehydrate using 5ml sterile saline peptone water (0.1% peptone with 0.85% NaCl). Recap securely & allow 5 minutes at room temperature for rehydration. Aseptically transfer 10 g of the PT item matrix from 2026MM01R1-3 to the sterile bag/bottle/flask & add 85 ml of diluent to each bag/bottle/flask. Empty the rehydrated vial to the diluent (prepared in step 3), ensuring thorough homogenization of the PT item. Incubate the prepared sample at 37°C for 2 hours and perform serial dilution: (101 – 105) & proceed with your routine analysis for Escherichia coli & Total plate count.
