Instructions to Participants

• Only trained and authorized personnel should handle PT items.
• Confirm sample integrity upon receipt and notify the PT Provider if damaged.
• Record date of receipt and store under the specified conditions until analysis.
• Initiate analysis within 48 hours of receipt.
• Treat PT items like routine samples and use standard methods unless otherwise instructed.

Revival Instructions

Vial 2025MF01R1-V1

1. Aseptically uncap the vial and rehydrate with 4 ml sterile saline peptone water (0.1% Peptone + 0.85% NaCl).
2. Securely recap and allow 5 minutes rehydration at room temperature.
3. Uncap container 2025MF01R1-1:
   • Transfer 25 g of sample matrix to Salmonella enrichment broth.
   • Transfer remaining 25 g to E. coli diluting fluid.
4. Homogenize thoroughly.
5. Transfer 2 ml rehydrated vial to Salmonella enrichment broth and remaining 2 ml to E. coli diluting fluid.
6. Perform routine analysis for: Salmonella and E. coli.

Vial 2025MF01R1-V2

1. Aseptically uncap the vial and rehydrate with 4 ml sterile saline peptone water (0.1% Peptone + 0.85% NaCl).
2. Securely recap and allow 5 minutes rehydration at room temperature.
3. Uncap container 2025MF01R1-2 and transfer 25 g of sample matrix to L. monocytogenes enrichment broth.
4. Homogenize thoroughly.
5. Transfer entire 4 ml rehydrated vial to L. monocytogenes enrichment broth.
6. Perform routine analysis for: L. monocytogenes.

Raw Data Submission Requirements

• Upload clear photographs of plates for all analytes tested.
• Upload photographs of biochemical tests performed.
• Accepted formats: JPG, PNG, PDF | Max size: 5 MB each.

Reporting of Results

For this PT round 2025MF01R1, you are required to report results for the following analytes:

• E. coli — Per g | IS 5887 (Part 1):1976
• Salmonella — Per 25 g | IS 5887 (Part 3):1999 / ISO 6579 (Part 1):2017
• L. monocytogenes — Per 25 g | IS 14988 (Part 1):2020 / BAM Chapter 10
